Open the file properties (right click on config.cfg -> properties) and see if "read-olny" is checked. If so, uncheck it and click "Apply".
        The config.cfg is located in /SteamApps/common/dota 2 beta/dota/cfg

You can try to delete you config.cfg. As far as I know dota 2 will create a new one once you start the game. But create a backup of the old config.cfg first.
All your settings will be set do default after deleting the config file!

            EDIT:
Do you have a custom autoexec.cfg which overwrites some of the settings you changed?

Please check if there is a file called "autoexec.cfg" in your /SteamApps/common/dota 2 beta/dota/cfg diretory.
As the name of the file tells you ... all commands inside this file will be executed automatically at game startup.

The other thing that can reset your video settings is having any of them set in your launch options. If you have -autoconfig or -dxlevel in your launch options that will force most of the settings to a specific value each time you launch.
